This quote represents his true calling. He finds himself feeling very fond of the river and having a desire to stay by it. Then hears his inner voice guiding him to stay there, learn from it, and love it, which led to his decision to stay with Vasudeva. With Vasudeva's guidance, he became not only a ferryman, but also a holy man who was finally at peace. The river led him to his destiny and helped him attain his long after nirvana, as it showed and told him many things.
The significance of this line is telling Siddhartha to stay by the river. He needs to love the river, stay by the river and learn from the river in order to continue on his path of enlightenment. The river teaches and shows him many things that no one else can, not even himself. The river symbolizes that time keeps going on and nothing ever stops.
